Background:

  • Plasminogen activator inhibitor-2 (PAI-2) inhibits plasminogen activators, serine proteases involved in brain plasticity and toxicity.
  • PAI-2 is primarily intracellular and may counteract apoptosis via an undefined protease.
  • The precise localization and induction of PAI-2 in the brain remain largely uncharacterized.

Purpose of the Study:

  • To investigate the localization of PAI-2 mRNA in the adult mouse brain under normal conditions.
  • To determine the changes in PAI-2 mRNA expression following kainate-induced excitation.
  • To elucidate the potential role of PAI-2 in brain activity and neuronal function.

Main Methods:

  • In situ hybridization was employed to detect PAI-2 mRNA.
  • Brain cryosections from normal adult mice were analyzed.
  • Sections from mice treated with kainate were also examined to assess induction patterns.

Main Results:

  • In normal adult mice, PAI-2 mRNA was detected specifically in the accumbens nucleus shell.
  • Following kainate injection, PAI-2 mRNA was rapidly induced (within 2 hours) in various brain regions.
  • Induced expression was observed in the neocortex, olfactory structures, striatum, limbic areas, and hippocampus.

Conclusions:

  • PAI-2 mRNA exhibits distinct localization in the normal brain, particularly in the accumbens nucleus.
  • Kainate-induced neuronal activity leads to a widespread and rapid induction of PAI-2 mRNA.
  • These findings suggest a role for PAI-2 in the accumbens nucleus and in activity-dependent processes within olfactory, striatal, and limbic systems.
