Abstract:
Lipid homeostasis in vertebrate cells is regulated by a family of membrane-bound transcription factors designated sterol regulatory element-binding proteins (SREBPs). SREBPs directly activate the expression of numerous genes dedicated to the synthesis and uptake of cholesterol, fatty acids, triglycerides, and phospholipids, together with the NADPH cofactor required to synthesize these molecules. In this review, we focus on the mechanism of SREBP trafficking and processing, as well as the molecular aspects of SREBPs in regulating cellular lipid homeostasis; furthermore, we discuss the involvement in the diseases relevant to the disturbance of lipid metabolism.
