Dentin resorption and cementum-like tissue formation by bone morphogenetic protein application

Bone morphogenetic protein-2 (BMP-2) at low doses stimulates cementum-like tissue formation on dentin, while high doses increase dentin resorption. Appropriate BMP-2 application may enhance periodontal regeneration.

- Rat root dentin blocks were treated with EDTA and exposed to varying concentrations of BMP-2 (0, 100, 400 µg/ml).
- Dentin blocks were implanted into rat palatal connective tissue.
- Histologic and histomorphometric analyses were performed at 2, 4, and 8 weeks post-surgery.

- BMP-2 demonstrated a dose-dependent increase in dentin resorption by osteoclastic cells.
- New cementum-like tissue formed on both resorbed and non-resorbed dentin surfaces in BMP-2 treated groups.
- Significantly greater dentin resorption occurred in BMP-2 treated groups compared to controls (p < 0.01).
- At 8 weeks, cementum-like tissue formation was observed in 41.8% of the 100 µg/ml BMP-2 group versus 16.2% in the 400 µg/ml group (p < 0.05).
Conclusions:
- High-dose BMP-2 stimulates dentin resorption, whereas low-dose BMP-2 induces cementum-like tissue formation.
- BMP-2 application at appropriate concentrations to dentin surfaces holds potential for enhancing periodontal regeneration.
