This study presents a novel microtransponder (MTP) based genotyping assay for detecting 50 cystic fibrosis (CF) mutations. The MTP assay demonstrates high accuracy for CF mutation detection.
Area of Science:
- Biotechnology
- Genetics
- Molecular Diagnostics
Background:
- Cystic Fibrosis (CF) is a genetic disorder.
- Accurate detection of CF mutations is crucial for diagnosis and management.
- Existing genotyping methods may have limitations in throughput or cost.
Purpose of the Study:
- To develop and evaluate a novel genotyping assay for detecting 50 cystic fibrosis (CF) mutations.
- To assess the performance of a microtransponder (MTP) based platform for CF genotyping.
Main Methods:
- Development of a multiplex assay utilizing microtransponder (MTP) technology.
- Amplification of 18 CF transmembrane regulator (CFTR) DNA fragments covering 50 mutations via multiplex PCR.
- Multiplex allele-specific primer extension (ASPE) reactions followed by hybridization to MTP-synthesized capture probes.
- Analysis using a custom automated fluorescence and radio frequency (RF) readout instrument.
Main Results:
- The assay was validated on 23 commercial DNA samples (internal study) and 32 DNA samples (external study).
- High accuracy was observed with correct call rates of 98.8% (internal) and 95.7% (external).
- Low false-positive (1.1% internal, 3.9% external) and false-negative (0.12% internal, 0.36% external) rates were reported.
Conclusions:
- The microtransponder (MTP)-based multiplex assay is a viable platform for cystic fibrosis (CF) genotyping.
- This technology offers a potentially efficient method for detecting multiple CF mutations.
- The assay demonstrates robust performance suitable for clinical applications.
