Summary

Local gene transfer of Interleukin-10 (IL-10) effectively reduces inflammatory responses to bacterial challenge. This approach shows promise for modulating inflammation in localized infections.

  • Local cytokine levels (IL-10, IFN-gamma, TNF-alpha, IL-1beta) were measured at 2 and 24 hours post-challenge.
  • Main Results:

    • Local IL-10 gene delivery successfully elevated intra-chamber IL-10 levels.
    • Significant attenuation of pro-inflammatory cytokines was observed: IFN-gamma and TNF-alpha at 2 hours, and IL-1beta at 24 hours.
    • Specific reductions included IFN-gamma (656 to 218 pg/mL), TNF-alpha (23 to 12.5 ng/mL), and IL-1beta (21.5 to 12.4 ng/mL).

    Conclusions:

    • Direct IL-10 gene transfer is a viable strategy for modulating local inflammatory responses.
    • This method shows potential for therapeutic intervention against localized bacterial infections like those caused by *P. gingivalis*.
