The origin recognition complex (ORC) interaction between Orc2 and Orc3 occurs in both the nucleus and cytoplasm, challenging previous assumptions about DNA replication initiation. These findings reveal new insights into the spatial regulation of genome duplication.
Area of Science:
- Cell Biology
- Molecular Biology
- Genetics
Background:
- Eukaryotic DNA replication initiation involves the origin recognition complex (ORC) binding to chromatin.
- The pre-replication complex (pre-RC) assembles at origins, enabling DNA replication.
- The MCM helicase is thought to translocate away from ORC to initiate DNA unwinding.
Purpose of the Study:
- To investigate the subcellular localization of Orc2-Orc3 interactions during pre-RC assembly.
- To explore the spatial dynamics of DNA replication initiation factors in live mammalian cells.
- To evaluate the utility of BRET and BiFC assays in studying genome duplication processes.
Main Methods:
- Utilized bioluminescence resonance energy transfer (BRET) and bimolecular fluorescence complementation (BiFC) assays in live mammalian cells.
- Assessed the interaction between Orc2 and Orc3, including with nuclear localization signal (NLS)-depleted Orc3.
- Examined the subcellular distribution of Orc2 and Orc3 relative to MCM proteins (Mcm3, Mcm6) and PCNA.
Main Results:
- Demonstrated that the Orc2-Orc3 interaction occurs in both the nucleus and cytoplasm, not exclusively in the nucleus.
- Confirmed a direct interaction between Orc2 and NLS-depleted Orc3 via BRET assays.
- Visualized the spatial distribution of key replication proteins, highlighting cellular complexity.
Conclusions:
- The spatial regulation of DNA replication initiation is more complex than previously understood.
- BRET and BiFC are valuable tools for studying the dynamic processes of genome duplication.
- Findings provide novel insights into the early events of DNA replication.
