[Prediction and analysis of the secreteomic in Trichoderma reesei]

Objective:
We studied the secretomics' properties of Trichoderma reesei, an important industrial microorganism used for cellulase production.
Methods:
We analyzed the amino acid sequences coded by 9997 ORFs in Trichoderma reesei genome with bioinformatics approaches, identified 294 possible secreted protein sequences, and classified them by functions. We also applied motif search methods to search key motifs in the function-unknown sequences and preliminary predicted their functions. Moreover, we analyzed the signal peptide sequences of the secreted proteins.
Results:
There were 188 hydrolytic enzymes in Trichoderma reesei's secretomics, including 114 glycosidases, 42 proteases, and 11 lipoidases. The glycosidases included 22 reported cellulases, and 15 chitinases, as well as 30 other protein sequences probably related to cellulose degradation. The homology of signal peptides of secreted proteins was low whereas sequences near the digesting site of signalase were conservative.
Conclusion:
This method gave insights into the whole secreted proteome of Trichoderma reesei and provided basis for further studies on secretomic features at a genome level.
