We developed a functional cell-based assay for high throughput screening (HTS) to discover metabotropic glutamate receptor 4 (mGluR4) modulators. This validated cell line enables efficient identification of novel mGluR4 agonists and antagonists.
Area of Science:
- Neuroscience
- Pharmacology
- Cell Biology
Background:
- Metabotropic glutamate receptor 4 (mGluR4) is a key target for neurological disorders.
- Identifying specific modulators requires robust and efficient screening assays.
Purpose of the Study:
- To develop a functional, cell-based high throughput screening (HTS) assay for metabotropic glutamate receptor 4 (mGluR4) modulators.
- To establish a stable recombinant cell line for screening mGluR4 agonists and antagonists.
Main Methods:
- Human mGluR4 cDNA was transfected into HEK-293 cells expressing promiscuous G-protein (Ga alpha15).
- A functional Ca2+ influx assay using Fluo Calcium Indicator was optimized.
- Assay performance was validated using known mGluR4 agonists and antagonists, and Z'-factor was calculated.
Main Results:
- A stable human mGluR4-expressing cell line was successfully generated and validated.
- The assay demonstrated high performance with Z'-factor values of 0.80 (96-well) and 0.65 (384-well).
- Rank order potency for agonists (L-AP4 > L-SOP > L-Glu) and antagonists (MSOP > MPPG) was established.
Conclusions:
- A robust and validated functional cell-based HTS assay for mGluR4 has been developed.
- This assay is suitable for screening large compound libraries to identify novel mGluR4 modulators.
- The established cell line provides a valuable tool for drug discovery targeting mGluR4.
