We created PICsL software to improve precursor ion assignment reliability in LC-MS analysis. This tool automatically corrects assignments, enhancing protein identification accuracy for mass spectrometry instruments.
Area of Science:
- Proteomics
- Mass Spectrometry
- Bioinformatics
Background:
- Accurate precursor ion assignment is crucial for reliable protein identification in LC-MS analysis.
- Existing computational methods for isotopic distribution calculation may have limitations in complex spectra.
Purpose of the Study:
- To develop a software program, PICsL, to enhance the reliability of precursor ion assignments in LC-MS analysis.
- To automate the correction of precursor ion assignments using ultra zoom scanning data.
Main Methods:
- Developed PICsL software for LTQ linear ion trap MS.
- Implemented a method to search for ions near theoretical mass values using MS/MS raw data and Mascot search results.
- Performed a second database search using regenerated peak list files for protein identification.
Main Results:
- PICsL software increases the reliability of precursor ion assignments.
- The software automatically corrects precursor ion assignments, mimicking manual spectral inspection.
- The method is applicable to low-resolution MS like LTQ and other MS instruments.
Conclusions:
- PICsL software offers a reliable and automated approach for precursor ion calibration in LC-MS.
- The developed method improves protein identification accuracy by enhancing precursor ion assignment.
- The software's broad applicability across various MS instruments makes it a valuable tool in proteomics research.
