Hybridoma Technology01:31

Hybridoma Technology

Hybridoma technology is used for the large-scale production of monoclonal antibodies. Monoclonal antibodies bind to only a single antigenic determinant or epitope. Such antibodies are used in research, diagnostics, and disease therapy. The hybridoma technology established in 1975 by Georges Köhler and Cesar Milstein was awarded the Nobel Prize in Medicine in 1984 for revolutionizing research and therapy.

Area of Science:

  • Immunology
  • Transplantation Science
  • Laboratory Medicine

Background:

  • Solid phase assays for human leukocyte antigen (HLA) antibody detection have advanced clinical organ transplantation.
  • Historical complement-dependent serological techniques had limitations in detecting pre-transplant immunization.

Purpose of the Study:

  • To review the advantages of fluorescent bead technology for HLA antibody determination.
  • To discuss technical issues and interpretation challenges associated with bead-based assays.

Main Methods:

  • Review of two generations of solid phase assays: ELISA and fluorescent bead technology.
  • Focus on fluorescent bead assays used with conventional flow cytometry or Luminex instrumentation.

Main Results:

  • Solid phase assays, particularly Luminex-based bead technology, have improved pre-transplant immunization detection.
  • These advancements have led to better matching and avoidance of organ transplant rejection.

Conclusions:

  • Fluorescent bead technology presents significant advantages for HLA antibody detection in transplantation.
  • Further research is needed to address technical performance and result interpretation for optimal clinical application.
