Genetic variations in cytochrome P4501A1 (CYP1A1) are linked to an increased risk of pterygium. Glutathione S-transferase M1 (GSTM1) polymorphisms showed no significant association with pterygium development.
Area of Science:
- Ophthalmology
- Genetics
- Environmental Health
Background:
- Polycyclic aromatic hydrocarbons (PAHs) are environmental pollutants metabolized by enzymes like cytochrome P4501A1 (CYP1A1) and glutathione S-transferase M1 (GSTM1).
- Benzo(a)pyrene (BaP) metabolites, such as BPDE-N2-dG adducts, can cause DNA damage and mutations, potentially contributing to diseases.
- Previous research linked CYP1A1 gene polymorphisms to BPDE-like DNA adduct levels in pterygium.
Purpose of the Study:
- To investigate the association between genetic polymorphisms of CYP1A1 and GSTM1 and the risk of developing pterygium.
- To determine if CYP1A1 and GSTM1 genetic variations are predictive markers for pterygium susceptibility.
Main Methods:
- Genotyping of CYP1A1 and GSTM1 was performed using DNA extracted from blood samples of 205 pterygium patients and 206 controls.
- Restriction fragment length polymorphism and polymerase chain reaction techniques were employed for mutation and genotype determination.
Main Results:
- A significant difference in CYP1A1 genotype distribution was observed between pterygium cases and controls (p=0.0161).
- CYP1A1 m2/m2 polymorphism showed a significantly increased odds ratio (1.647, p=0.006) for pterygium compared to the wild-type.
- No significant association was found between GSTM1 polymorphisms and pterygium risk (p=1.000).
Conclusions:
- CYP1A1 genetic polymorphism is significantly correlated with pterygium.
- CYP1A1 variations may serve as a potential biomarker for predicting pterygium susceptibility.
- GSTM1 polymorphisms do not appear to influence pterygium risk in this population.
