Obesity-related inflammation in fat tissue involves the calcium sensing receptor (CaSR). CaSR activation increases inflammatory factors, partly via nuclear factor kappa B (NFκB), suggesting therapeutic potential.
Area of Science:
- Biochemistry
- Cell Biology
- Endocrinology
Background:
- Adipose tissue inflammation is linked to obesity's metabolic and cardiovascular issues.
- The calcium sensing receptor (CaSR) is present in human fat cells and upregulated during obesity-related inflammation.
Purpose of the Study:
- To investigate the role of CaSR in promoting inflammatory factor expression in adipose tissue.
- To explore the potential of CaSR as a therapeutic target for obesity-induced adipose tissue dysfunction.
Main Methods:
- Activation of CaSR using cinacalcet in adipose tissue and LS14 cells.
- Measurement of inflammatory cytokine and chemokine expression (IL6, IL1β, TNFα, CCL2).
- Inhibition of nuclear factor kappa B (NFκB) pathway using SN50.
Main Results:
- CaSR activation significantly increased the expression of proinflammatory cytokines IL6, IL1β, TNFα, and chemokine CCL2.
- The observed increase in inflammatory factors was partially reversed by NFκB inhibition.
- These findings indicate CaSR activation elevates cytokine and chemokine production, partly mediated by NFκB.
Conclusions:
- CaSR plays a significant role in obesity-induced adipose tissue dysfunction.
- CaSR activation contributes to adipose tissue inflammation through NFκB signaling.
- Targeting CaSR presents a potential therapeutic strategy for managing obesity-related complications.
