T Cell Types and Functions01:24

T Cell Types and Functions

When T cells with CD4 markers are activated, they give rise to two types of effector cells: helper T cells and regulatory T cells. Meanwhile, T cells with CD8 markers differentiate into effector cytotoxic T cells. The differentiation of CD4 T cells into helper T cell subsets, such as Th1, Th2, and Th17 cells, is dependent on the antigen type, antigen-presenting cell, and regulatory cytokines.

T Cell Activation and Clonal Selection

T cells are integral to our adaptive immune system, recognizing and effectively responding to foreign antigens. T cell activation and clonal selection are pivotal in orchestrating this immune response. This article elucidates these mechanisms, detailing the roles of cluster of differentiation (CD) markers, major histocompatibility complex (MHC) molecules, costimulatory signals, and the process of clonal selection.
Naive T cells that have not yet encountered an antigen express two primary CD...

Summary

Interleukin-27 (IL-27) induces IL-10 production in Tr1 cells via STAT3 and Egr-2, independent of c-Maf/Ahr. This reveals plasticity in CD4(+) T cell differentiation pathways.

Area of Science:

  • Immunology
  • Cell Biology
  • Molecular Biology

Background:

  • Tr1 cells are regulatory CD4(+) T cells that secrete IL-10.
  • Interleukin-27 (IL-27) promotes Tr1 cell differentiation and IL-10 production, partly via c-Maf and Ahr transcription factors.

Purpose of the Study:

  • To investigate an alternative IL-27-regulated transcriptional network for IL-10 induction in Tr1 cells.
  • To elucidate the molecular mechanisms underlying IL-10 production by Tr1 cells.

Main Methods:

  • Analysis of transcription factor activation (STAT3, Egr-2, c-Maf, Ahr, Blimp1).
  • Investigation of IL-10 gene induction in Tr1 cells.
  • Study of IL-27 signaling pathways.

Main Results:

  • IL-27 activates STAT3 and Egr-2 transcription factors in a Blimp1-dependent manner to induce IL-10.
  • This STAT3/Egr-2 pathway is independent of c-Maf and Ahr.
  • IL-10 induction in Tr1 cells is context-dependent, influenced by cytokine environment and molecular factors.
  • Conclusions:

    • IL-27 utilizes a distinct transcriptional network involving STAT3 and Egr-2 for IL-10 induction in Tr1 cells.
    • Tr1 cells exhibit plasticity, potentially representing a developmental endpoint rather than a distinct lineage.
    • Findings highlight the complex regulation of T helper cell differentiation and function.
