Dual phosphoinositide 3-kinase/mammalian target of rapamycin (PI3K/mTOR) inhibitors are emerging as promising anticancer agents. This review highlights recent advances in small molecule dual PI3K/mTOR inhibitors for cancer therapy.
Area of Science:
- Molecular Biology
- Oncology
- Medicinal Chemistry
Background:
- The phosphoinositide 3-kinases (PI3Ks) pathway, including protein kinase B (Akt) and mammalian target of rapamycin (mTOR), is crucial for cell growth, survival, and cancer progression.
- Dysregulation of the PI3K/Akt/mTOR pathway is implicated in various cancers, making it a significant target for anticancer drug development.
- Targeting both PI3K and mTOR simultaneously offers a potential strategy to enhance therapeutic efficacy by suppressing cancer cell proliferation and inducing apoptosis.
Purpose of the Study:
- To review the recent developments in small molecule dual PI3K/mTOR inhibitors over the past decade.
- To provide an overview of the structural features, biological activities, and structure-activity relationships (SARs) of these inhibitors.
Main Methods:
- Literature review focusing on small molecule dual PI3K/mTOR inhibitors published in the last 10 years.
- Analysis of structural characteristics, biological data, and SARs of identified inhibitors.
Main Results:
- A diverse range of small molecule dual PI3K/mTOR inhibitors have been developed.
- These inhibitors exhibit varying structural scaffolds and demonstrate significant biological activities in preclinical cancer models.
- Structure-activity relationship studies have provided insights into optimizing inhibitor design for enhanced potency and selectivity.
Conclusions:
- Dual PI3K/mTOR inhibitors represent a promising class of anticancer therapeutics.
- Continued research into their structural features and SARs is crucial for the development of effective cancer treatments.
- These inhibitors hold potential for preventing cancer cell proliferation and inducing apoptosis by effectively suppressing the PI3K/Akt/mTOR pathway.
