A new 34-gene signature, ClearCode34, classifies clear cell renal cell carcinoma (ccRCC) into good (ccA) and poor (ccB) risk subtypes. This molecular biomarker improves risk prediction for nonmetastatic ccRCC patients, enhancing survival outcome assessments.
Area of Science:
- Oncology
- Genomics
- Biomarker Discovery
Background:
- Gene expression signatures are crucial for cancer subtyping and predicting outcomes.
- Current kidney cancer diagnostics lack clinically applicable molecular signatures.
Purpose of the Study:
- Develop a novel gene expression biomarker for clear cell renal cell carcinoma (ccRCC) risk stratification.
- Classify ccRCC into good-risk (ccA) and poor-risk (ccB) subtypes for improved clinical management.
Main Methods:
- A 34-gene classifier (ClearCode34) was developed using ccRCC sample standards.
- The classifier was validated on RNA-sequencing data from The Cancer Genome Atlas (TCGA) and clinical formalin-fixed samples.
Main Results:
- ClearCode34 effectively classified ccRCC tumors into ccA and ccB subtypes.
- Subtype classification showed significant associations with recurrence-free survival, cancer-specific survival, and overall survival (p<0.01).
- The 34-gene signature demonstrated prognostic value comparable to tumor stage and grade.
Conclusions:
- The ClearCode34 gene signature provides robust prognostic stratification for nonmetastatic ccRCC.
- An integrated molecular and clinical model using ClearCode34 improves prediction of patient outcomes.
- This biomarker enhances risk assessment for recurrence and mortality in ccRCC patients.

When the function of certain critical genes, especially those involved in cell cycle regulation and cell growth signaling cascades, gets disrupted, it upsets the cell cycle progression. Such cells with unchecked cell cycles start proliferating uncontrollably and eventually develop into tumors.

Tumor Progression
Colon cancer is one of the best-documented examples of tumor progression. Early mutation in the APC gene in colon cells causes a small growth on the colon wall called a polyp. With time, this polyp grows into a benign, pre-cancerous tumor. Further...
