We developed a simple DNA-linker method for bioconjugating upconversion nanoparticles (UCNPs) with proteins. This rapid technique enhances UCNP stability and specificity for biotechnological applications like immunoassays.
Area of Science:
- Nanotechnology
- Bioconjugation Chemistry
- Biomedical Diagnostics
Background:
- Upconversion nanoparticles (UCNPs) are promising luminescent probes for biotechnology, but effective bioconjugation is challenging.
- Existing bioconjugation methods are multi-step, leading to poor probe stability, specificity, and reproducibility.
- Achieving stable, specific protein attachment to UCNPs is critical for applications like molecular recognition and imaging.
Purpose of the Study:
- To develop a simple, rapid, and efficient method for bioconjugating proteins to upconversion nanoparticles.
- To improve the stability, specificity, and aqueous dispersibility of functionalized upconversion nanoparticles.
- To demonstrate the utility of the developed bioconjugation strategy in biological assays.
Main Methods:
- A one-step ligand exchange protocol using DNA as a linker molecule for protein conjugation to UCNPs.
- Proteins are pre-conjugated to the 3' end of synthetic DNA, utilizing the DNA's phosphate groups for nanoparticle surface interaction.
- The method facilitates the transfer of UCNPs from organic solvents to aqueous phases, rendering them hydrophilic.
Main Results:
- Successfully created robust DNA-protein conjugates for UCNP functionalization.
- Demonstrated efficient transfer of UCNPs into aqueous solutions, resulting in stable and hydrophilic nanoparticles.
- Functionalized UCNPs with horseradish peroxidase (HRP) and streptavidin (SA) for successful catalytic colorimetric and immunoassay applications.
Conclusions:
- The novel DNA-mediated one-step ligand exchange offers a simple and rapid bioconjugation strategy for upconversion nanoparticles.
- This method significantly enhances nanoparticle stability, specificity, and aqueous compatibility for biotechnological applications.
- The functionalized UCNPs show great potential for sensitive biomarker detection and immunoassays.
