Hematopoietic cells can transform into fibroblast-like cells using macrophage colony-stimulating factor (M-CSF). These cells can then differentiate into multiple cell types, offering a new method for generating multipotent stem cells.
Area of Science:
- Cell Biology
- Stem Cell Research
- Hematopoiesis
Background:
- Hematopoietic cells possess potential for trans-differentiation into non-lymphohematopoietic lineages.
- Mechanisms and factors driving hematopoietic cell trans-differentiation are largely unknown.
Purpose of the Study:
- To investigate the factors and mechanisms inducing hematopoietic cell trans-differentiation.
- To identify a method for generating multipotent stem cells from hematopoietic cells.
Main Methods:
- Utilized an in vitro culture system with conditioned fibroblast medium.
- Purified active factors from conditioned medium using molecular weight cut-offs, ammonium sulfate precipitation, and fast protein liquid chromatography (FPLC).
- Identified the active factor using mass spectrometry and analyzed induced cell markers and differentiation potential.
Main Results:
- Conditioned fibroblast medium induced hematopoietic cells to become fibroblast-like cells (FLCs).
- Macrophage colony-stimulating factor (M-CSF) was identified as the key inducing protein.
- FLCs expressed stem cell markers (SSEA-1, OCT3/4, NANOG, SOX2) and differentiated into three germ layers.
Conclusions:
- M-CSF induces hematopoietic cells to dedifferentiate into multipotent stem cells.
- This process involves a tyrosine kinase signaling pathway.
- Provides a novel, simple method for generating multipotent stem cells for potential clinical applications.
