Area of Science:

  • Forensic Science
  • Molecular Biology
  • Genetics

Background:

  • Accurate quantification of human DNA is essential for downstream Short Tandem Repeat (STR) DNA analysis.
  • Existing commercial methods offer limited insight into DNA sample quality, such as degradation or PCR inhibitors.
  • Federal quality assurance standards mandate DNA quantification for forensic evidence.

Purpose of the Study:

  • To perform a developmental validation of the PowerQuant System for forensic DNA quantification.
  • To evaluate the PowerQuant System's ability to quantify total human and male DNA concentrations.
  • To assess the system's capacity for detecting DNA degradation and PCR inhibitors.

Main Methods:

  • Developmental validation of the PowerQuant System following SWGDAM Guidelines.
  • Assay evaluation included specificity, sensitivity, precision, and accuracy.
  • Testing for the detection of degraded DNA and PCR inhibitors using specific targets.

Main Results:

  • The PowerQuant System accurately quantifies total human and total human male DNA.
  • The system successfully identifies the presence of degraded DNA.
  • Internal PCR controls (IPC) effectively indicate the presence of PCR inhibitors.

Conclusions:

  • The PowerQuant System provides comprehensive DNA quantification and quality assessment.
  • Degradation and PCR inhibition data aid in selecting optimal forensic workflows.
  • This system offers valuable information for downstream STR DNA analysis interpretation.
