Two high-performance liquid chromatography (HPLC) systems were developed for simultaneously determining lipid hydroperoxides. These methods accurately quantify hydroperoxides in various lipids at the picomole level with good reproducibility.
Area of Science:
- Analytical Chemistry
- Biochemistry
- Lipidomics
Background:
- Lipid hydroperoxides are critical biomarkers of oxidative stress.
- Accurate quantification of lipid hydroperoxides is essential for understanding lipid peroxidation.
- Existing methods may lack sensitivity or specificity for simultaneous determination.
Purpose of the Study:
- To develop and validate two novel HPLC-post column systems for simultaneous lipid hydroperoxide determination.
- To enable sensitive and reproducible quantification of various lipid hydroperoxide species.
Main Methods:
- Development of two HPLC-post column systems: a conventional-size and a semi-micro system.
- Utilized gradient elution for optimal separation.
- Employed post-column detection with diphenyl-1-pyrenylphosphine for sensitive detection.
Main Results:
- Successful simultaneous determination of lipid hydroperoxides from free fatty acids, phosphatidylcholines, triacylglycerols, and cholesterol esters.
- Achieved quantification at the picomole (pmol) level.
- Demonstrated good reproducibility for the developed methods.
Conclusions:
- The developed HPLC-post column systems offer a robust and sensitive approach for simultaneous lipid hydroperoxide analysis.
- These methods are valuable tools for research in oxidative stress and lipid metabolism.
- The semi-micro system provides a potentially more efficient option for high-throughput analysis.
