  • Materials Science

Background:

  • Liposomes are versatile drug delivery systems.
  • Antibody functionalization of liposomes enhances targeting capabilities.
  • Avidin-biotin system offers strong molecular interactions.

Purpose of the Study:

  • To develop antibody-derivatized liposomes for immunoassays.
  • To investigate the binding activity of avidin on liposome surfaces.
  • To optimize liposome derivatization for immunospecific aggregation.

Main Methods:

  • Liposomes were derivatized with antibodies using avidin-biotin crosslinking.
  • Avidin's biotin-binding activity was compared in solution and on liposomes.
  • Fluorescence spectroscopy and light scattering were used for characterization.

Related Experiment Videos

  • Optimization of biotinylated phospholipid, avidin, and antibody ratios was performed.
  • Main Results:

    • Avidin bound to liposomes retained full biotin-binding activity.
    • Stable liposomes were produced with up to 1.7 nmol antibody/µmol lipid.
    • Derivatized liposomes showed high reactivity for immunospecific aggregation.
    • A linear increase in light scattering was observed with 1-10 pmol of antigen.

    Conclusions:

    • Liposomes containing biotinylated phospholipids are effective generic reagents for immunoassays.
    • The developed method allows for stable antibody conjugation to liposomes.
    • These liposomes demonstrate sensitive detection of multivalent antigens.
