Mitochondria in heart cells with a RyR2 mutation extend more nanotunnels, which are thin connections between mitochondria. These nanotunnels facilitate slower exchanges of mitochondrial matrix contents compared to direct contact.
Area of Science:
- Cell Biology
- Mitochondrial Dynamics
- Cardiovascular Research
Background:
- Mitochondrial matrix content exchange is crucial for mitochondrial maintenance.
- Cardiac mitochondria communicate via direct contact and long-distance nanotunnels.
- RyR2 mutations are linked to catecholaminergic polymorphic ventricular tachycardia and can cause Ca2+ imbalance.
Purpose of the Study:
- To investigate the role of mitochondrial nanotunnels in cardiac myocytes with a RyR2 mutation.
- To characterize the ultrastructure and 3D distribution of nanotunnels.
- To compare the kinetics of matrix content exchange via nanotunnels versus direct contact.
Main Methods:
- Utilized heterozygous mice with the RyR2 A4860G mutation (RyR2A4860G+/-).
- Employed electron tomography to reconstruct nanotunnel ultrastructure and 3D distribution.
- Assessed intermitochondrial matrix content exchange using fluorescent proteins (mtDsRed, mtPA-GFP) in isolated cardiomyocytes via confocal microscopy.
Main Results:
- RyR2A4860G+/- cardiomyocytes exhibit a significantly increased frequency of mitochondrial nanotunnel extensions.
- Electron tomography provided detailed ultrastructural analysis and 3D mapping of these nanotunnels.
- Confocal microscopy revealed that communication via nanotunnels is slower than direct mitochondrial contact.
Conclusions:
- A specific RyR2 mutation promotes increased mitochondrial nanotunnel formation in cardiac myocytes.
- Mitochondrial nanotunnels represent a distinct pathway for inter-mitochondrial communication.
- Nanotunnel-mediated exchange of matrix contents is a slower process compared to direct mitochondrial interactions.

Sorting of outer membrane proteins:
Mitochondrial outer membrane proteins are of two types: the transmembrane, beta-barrel porins, and the membrane-anchored, alpha-helical proteins. Beta-barrel porin precursors are translocated by the TOM complex and inserted into the outer mitochondrial membrane by the SAM complex. In contrast,...
