Summary
We developed a novel biosensor using graphene oxide (GO) modified pencil graphite electrodes (PGEs) for sensitive detection of miRNA-34a, a biomarker linked to Alzheimer disease.
Area of Science:
- Electrochemistry
- Biosensors
- Nanomaterials
Background:
- MicroRNAs (miRNAs) play crucial roles in cellular processes and disease development.
- miRNA-34a is implicated in the pathogenesis of Alzheimer disease.
- Development of sensitive and selective detection methods for miRNAs is essential.
Purpose of the Study:
- To develop and characterize a novel electrochemical biosensor for the detection of miRNA-34a.
- To investigate the application of graphene oxide (GO) modified pencil graphite electrodes (PGEs) for miRNA monitoring.
- To explore DNA-RNA hybridization for specific miRNA recognition.
Main Methods:
- Modification of pencil graphite electrodes (PGEs) with graphene oxide (GO).
- Electrochemical characterization using cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and scanning electron microscopy (SEM).
- Voltammetric monitoring of miRNA-34a using differential pulse voltammetry (DPV) with DNA-RNA hybridization.
Main Results:
- Successful modification of PGEs with GO was confirmed.
- The developed CA/GO/PGE biosensor demonstrated sensitive detection of miRNA-34a.
- Optimization of GO concentration, DNA probe concentration, and miRNA-34a concentration was achieved.
- The biosensor exhibited good selectivity for miRNA-34a.
Conclusions:
- Graphene oxide modified pencil graphite electrodes offer a promising platform for electrochemical miRNA detection.
- The developed biosensor provides a sensitive and selective method for monitoring miRNA-34a.
- This approach holds potential for early diagnosis and monitoring of Alzheimer disease.
