Potential association between ITPKC genetic variations and Hirschsprung disease

Genetic variants in inositol-trisphosphate 3-kinase C (ITPKC) may be associated with Hirschsprung disease (HSCR), a congenital disorder affecting enteric neuron development. Further research is needed to confirm these findings in larger patient cohorts.

Background:
- Hirschsprung disease (HSCR) is a congenital disorder causing intestinal obstruction due to absent enteric neurons.
- Genome-wide association studies (GWAS) previously indicated a link between HSCR and loci of inositol-trisphosphate 3-kinase C (ITPKC).

Main Results:
- Identified several SNPs and a haplotype significantly associated with HSCR (minimum P=0.004).
- rs76785336 variant showed altered mRNA folding structure.
- rs2607420 was predicted to create a new splice site.
Conclusions:
- Genetic variants in ITPKC show potential association with Hirschsprung disease susceptibility.
- These findings contribute to understanding the genetic basis of enteric nervous system development disorders.
- Further functional evaluations and replication studies are warranted.
