Area of Science:

  • Endocrinology
  • Hepatic Metabolism
  • Pharmacology

Background:

  • Somatostatin is used to inhibit pancreatic hormone secretion.
  • Previous studies suggested but did not confirm direct hepatic effects of somatostatin.

Purpose of the Study:

  • To investigate the direct effects of somatostatin on hepatic insulin extraction (HIE) and hepatic glucose production (HGP).

Main Methods:

  • Utilized anesthetized dogs, infusing insulin alone and with somatostatin.
  • Measured portal and hepatic artery blood flow electromagnetically.
  • Collected blood samples from multiple sites to determine HIE and HGP.

Main Results:

  • Somatostatin significantly increased hepatic insulin extraction (72.0% vs. 64.0%).

Related Experiment Videos

  • Somatostatin decreased hepatic glucose production (1.71% vs. 3.72%).
  • Conclusions:

    • Somatostatin administration increases the liver's extraction of exogenous insulin.
    • This increased insulin extraction is associated with a reduction in hepatic glucose production.
