Improving Translational Accuracy02:07

Improving Translational Accuracy

15.1K
Base complementarity between the three base pairs of mRNA codon and the tRNA anticodon is not a failsafe mechanism. Inaccuracies can range from a single mismatch to no correct base pairing at all. The free energy difference between the correct and nearly correct base pairs can be as small as 3 kcal/ mol. With complementarity being the only proofreading step, the estimated error frequency would be one wrong amino acid in every 100 amino acids incorporated. However, error frequencies observed in...

Nonsense-mediated mRNA Decay02:27

Nonsense-mediated mRNA Decay

11.9K
The Upf proteins that carry out nonsense-mediated decay (NMD) are found in all eukaryotic organisms, including humans. Each protein has an individual role, but they need to work in collaboration. Upf1 is an ATP-dependent RNA helicase that unwinds the RNA helix. Because Upf1 can unwind any RNA, Upf2 and Upf3 are required to help Upf1 discriminate between nonsense and normal mRNAs.
Usually, Upf3 binds to an Exon Junction Complex (EJC) at mRNA splice sites. If a ribosome fully translates the mRNA,...

piRNA - Piwi-interacting RNAs02:57

piRNA - Piwi-interacting RNAs

7.7K
PIWI-interacting RNAs, or piRNAs, are the most abundant short non-coding RNAs. More than 20,000 genes have been found in humans that code for piRNAs while only 2000 genes have been found for miRNAs. piRNAs can act at the transcriptional and post-transcriptional levels and have a vital role in silencing transposable elements present in germ cells. They are also involved in epigenetic silencing and activation. Previously, they were thought to function only in germ cells but new evidence suggests...

    Summary
    This summary is machine-generated.

Premature adrenarche, early onset of adrenal androgen production, requires excluding other conditions. It may predict future hormonal issues, necessitating observation and lifestyle management.

    Area of Science:

    • Pediatric Endocrinology
    • Adolescent Medicine
    • Reproductive Health

    Background:

    • Adrenarche is the onset of adrenal cortex secretion of androgen precursors like dehydroepiandrosterone (DHEA).
    • Pubarche, characterized by pubic hair, body odor, and acne, results from androgen conversion.
    • Premature adrenarche occurs before age 8 in girls and 9 in boys.

    Purpose of the Study:

    • To define premature adrenarche and its diagnostic considerations.
    • To explore associations between premature adrenarche and early life factors.
    • To outline management strategies for premature adrenarche.

    Main Methods:

    • Diagnosis of premature adrenarche is one of exclusion.
    • Rule out precocious puberty, congenital adrenal hyperplasia, and androgen excess.
    • Assess for history of small for gestational age birth with rapid weight gain or obesity.

    Main Results:

    • Premature adrenarche may be linked to specific infant growth patterns.
    • It can indicate potential for functional ovarian hyperandrogenism later in life.
    • Dehydroepiandrosterone sulfate serves as a marker for adrenal androgen activity.

    Conclusions:

    • Management focuses on observation and lifestyle adjustments.
    • Monitoring for persistent androgen excess and insulin resistance is crucial.
    • Premature adrenarche requires careful evaluation to rule out other endocrine disorders.
