    Background:

    • Ta1 antigen is a 105,000 dalton protein expressed on a small fraction of resting human peripheral blood T cells.
    • It is strongly expressed on T cell clones and activated T cells, irrespective of cell cycle, unlike some growth factor receptors.

    Purpose of the Study:

    • To investigate the function of the Ta1 antigen in human T lymphocytes.
    • To determine if Ta1 expression correlates with specific T cell responses, such as anamnestic responses and autologous mixed lymphocyte reactions.

    Main Methods:

    • Separation of human peripheral blood T lymphocytes into Ta1-enriched and Ta1-depleted subpopulations.
    • Assessment of anamnestic responses to recall antigens (tetanus toxoid, mumps).
    • Evaluation of responses to mitogen (PHA) and autologous mixed lymphocyte response.
    • Analysis of Ig synthesis induction and suppressor cell activity.

    Main Results:

    • The Ta1-enriched T cell subpopulation (10-15% of lymphocytes) contained most of the anamnestic response to recall antigens.
    • Both Ta1-enriched and -depleted cells responded equally to PHA, indicating Ta1 is not required for general mitogenic activation.
    • The autologous mixed lymphocyte response was higher in Ta1-enriched cells, and Ta1- cells did not exhibit suppressor activity.

    Conclusions:

    • The Ta1 antigen serves as a marker for previously activated T cells in peripheral blood.
    • The Ta1+ T cell subpopulation appears to encompass T memory cells, crucial for adaptive immunity.
    • Ta1 expression is associated with antigen-specific memory but not general T cell activation or suppression.
