We developed a low-cost, 3D microfluidic chip using photonic crystal beads for multiplex bioassays. This device enables sensitive biomarker detection with reduced background noise, showing promise for clinical diagnostics.
Area of Science:
- Biomedical Engineering
- Analytical Chemistry
- Materials Science
Background:
- Point-of-care testing (POCT) is crucial for clinical, environmental, and biomedical applications.
- Multiplex bioassays require sensitive and convenient biomarker detection methods.
- Photonic crystal beads (PCBs) offer self-coding capabilities for bioassay integration.
Purpose of the Study:
- To develop a cost-effective, mass-producible 3D microfluidic chip for multiplex bioassays using PCBs.
- To evaluate the chip's performance in terms of sensitivity, background reduction, and multiplexing capability.
Main Methods:
- Fabrication of a 3D microfluidic chip using polyethylene terephthalate, polymethyl methacrylate, a Ni square mesh grid, and double-sided tape.
- Immobilization of probes onto PCBs for target molecule capture in a flow-through manner.
- Assessment of chip performance using anti-human alpha fetoprotein (AFP) and simultaneous detection of multiple biomarkers.
Main Results:
- The 3D microfluidic chip required less washing and exhibited reduced background compared to flow-over designs.
- The limit of detection (LOD) for anti-human AFP was 18.92 ng/mL, suitable for clinical use.
- Simultaneous detection of human immunoglobulin G, carcinoembryonic antigen, and AFP was demonstrated.
Conclusions:
- The developed 3D microfluidic chip based on PCBs is a promising platform for sensitive and multiplexed point-of-care testing.
- The low-cost, mass-producible design facilitates broad applications in clinical diagnostics and biomedical research.

Ionic Crystal Structures
Most monatomic ions behave as charged spheres, and their attraction for ions of opposite charge is the same in every direction. Consequently, stable structures for ionic compounds result (1) when ions of one charge are surrounded by as many ions as possible of the opposite...
Crystal Field Theory - Octahedral Complexes
To explain the observed behavior of transition metal complexes (such as colors), a model involving electrostatic interactions between the electrons from the ligands and the electrons in the unhybridized d orbitals of the central metal atom has been developed. This electrostatic model is crystal field theory (CFT). It helps to understand, interpret, and predict the colors, magnetic behavior, and some structures of coordination compounds of transition metals.
CFT focuses on...
Crystal Growth: Principles of Crystallization
Initiating crystallization involves manipulating the concentration of the solute and the temperature of the solution. Since crystal growth occurs when the ratio of concentration and solubility of the solute in the solvent...
Crystal Field Theory - Tetrahedral and Square Planar Complexes
Crystal field theory (CFT) is applicable to molecules in geometries other than octahedral. In octahedral complexes, the lobes of the dx2−y2 and dz2 orbitals point directly at the ligands. For tetrahedral complexes, the d orbitals remain in place, but with only four ligands located between the axes. None of the orbitals points directly at the tetrahedral ligands. However, the dx2−y2 and dz2 orbitals (along the Cartesian axes) overlap with the ligands less than the dxy,...
