  • Adseverin (Ads) is a Ca2+-dependent actin-binding protein.
  • High Ads expression is observed in gastric, prostate, and bladder cancers.
  • The role of Ads in cancer cell actin remodeling, extension formation, and matrix invasion remains unclear.

Purpose of the Study:

  • To investigate the role of Adseverin (Ads) in subcortical actin remodeling.
  • To determine Ads' contribution to cell extension formation and matrix invasion in cancer cells.

Main Methods:

  • CRISPR/Cas9 was used to generate Adseverin wildtype and Ads-null MCF7 breast cancer cells.
  • Cell morphology, extension formation, and matrix degradation were compared between wildtype and Ads-null cells.
  • Cell migration and collagen degradation assays were performed on 2D and 3D matrices.

Main Results:

  • Ads-null cells showed significantly reduced cell extension formation (37% on fibronectin) and a more circular morphology.
  • Reconstitution of Ads restored cell extension formation in Ads-null cells.
  • Ads deletion reduced cell invasion through Transwell membranes (~40%) and collagen degradation (including MT1-MMP expression and phagocytosis).

Conclusions:

  • Adseverin plays a crucial role in cell extension formation and collagen degradation in MCF7 breast cancer cells.
  • These Ads-dependent processes may influence cancer cell matrix invasion and metastasis.
  • Targeting Adseverin could be a potential therapeutic strategy for certain cancers.
