We developed a good manufacturing practice (GMP)-compliant protocol to derive long-term neuroepithelial stem cell progenitors (lt-NES) from clinical-grade stem cells. This adaptable protocol supports the development of neurological therapies and regenerative medicine.
Area of Science:
- Stem Cell Biology
- Neuroscience
- Regenerative Medicine
Background:
- Clinical translation of human pluripotent stem cells requires safe and controlled differentiation protocols.
- Adapting research-grade reagents to Good Manufacturing Practice (GMP) standards is essential but challenging.
- Limited published protocols exist to guide this critical translational step.
Purpose of the Study:
- To adapt existing research protocols for deriving and differentiating long-term neuroepithelial stem cell progenitors (lt-NES) using GMP-grade reagents.
- To establish a robust and reproducible protocol for clinical applications.
Main Methods:
- Adapted research-grade protocols for lt-NES derivation and differentiation to GMP-grade reagents.
- Screened protocol robustness using six clinical-grade human embryonic stem cell (hESC) lines.
- Validated the derivation and differentiation of lt-NES under GMP conditions.
Main Results:
- Successfully derived lt-NES that are multipotent, bankable, and karyotypically stable.
- Demonstrated robust and reproducible differentiation of clinical-grade hESCs into lt-NES.
- Showcased high neurogenic potential and subtype differentiation capacity of GMP-derived lt-NES.
Conclusions:
- Reported the feasibility of deriving and differentiating clinical-grade hESCs into lt-NES under GMP conditions.
- The developed protocol is a flexible tool for accelerating the clinical translation of pluripotent stem cells.
- This work facilitates the development of novel neurological therapies and regenerative medicine strategies.
