Summary

This study developed a novel biofunctionalized titanium dioxide surface using bisphosphonate linkers to enhance implant osseointegration. The modified surface significantly improved pre-osteoblast adhesion and gene expression, paving the way for advanced dental implants.

Background:

Endosseous implants require improved biocompatibility and osteoinductivity for better performance.
Bisphosphonate molecules offer a strategy to link titanium dioxide surfaces with proteins, potentially aiding osseointegration.

Purpose of the Study:

To develop and evaluate a novel biofunctionalized titanium dioxide (TiO2) surface using sodium alendronate (ALN) as a linker and bovine serum albumin as the protein.
To assess the impact of this functionalization on surface properties, pre-osteoblast adhesion, and related gene expression.

Main Methods:

Surface functionalization of TiO2 with sodium alendronate and bovine serum albumin.
Physicochemical characterization using contact angle and surface roughness measurements.
Biocompatibility assessment via pre-osteoblast adhesion assays and analysis of cell-adhesion-related gene expression (Bsp, Bglap2) and matrix metalloproteinase activity.

Main Results:

Efficient surface functionalization was confirmed by physicochemical analyses.
A significant increase (up to 100-fold) in pre-osteoblast adhesion was observed on the modified surfaces compared to controls.
Enhanced expression of Bsp and Bglap2 genes (approximately sixfold change) was noted in the presence of ALN, indicating improved osteogenic potential.
Extracellular matrix remodeling was identified as crucial for early-stage cell adhesion.

Conclusions:

Bisphosphonate-biofunctionalized TiO2 surfaces demonstrate enhanced biocompatibility and promote cell adhesion.
This novel approach holds promise for improving the performance of endosseous implants.
Further preclinical in vivo studies are warranted to validate these findings.
