Introduction to Fibroblasts

3.6K
Rudolph Virchow discovered spindle-shaped cells called fibroblasts in 1858. Inactive fibroblasts, called fibrocytes, become activated by various stimuli, such as growth factors and inflammatory cytokines. Activated fibroblasts play a crucial role in wound healing, inflammation, formation of new blood vessels, and cancer progression. Uncontrolled activation of fibroblasts results in fibrosis, the excess deposition of fibrous tissue, which can lead to scarring and affect normal organs. This...

Area of Science:

  • Biomedical Engineering
  • Dermatology
  • Photomedicine

Background:

  • Keloids result from excessive collagen synthesis and fibroblast activity, with no standard treatments preventing recurrence.
  • Physical therapies, including PhotoBioModulation (PBM), are being explored for keloid management.

Purpose of the Study:

  • To investigate the effects of blue LED light irradiation on human keloid and perilesional fibroblasts.
  • To assess PBM's potential as an innovative treatment for keloids and hypertrophic scars.

Main Methods:

  • Human fibroblasts from keloids and perilesional tissue were irradiated with blue LED light (410-430 nm) at various doses.
  • Cell metabolism, proliferation, viability, Cytochrome C oxidase activity, and ionic membrane currents were analyzed.

Main Results:

  • Blue LED light modulated fibroblast metabolism and proliferation in a dose-dependent manner, with effects lasting up to 48 hours.
  • High light doses reduced fibroblast viability, with faster effects on keloid fibroblasts.
  • A medium dose enhanced voltage-dependent outward currents in fibroblasts.

Conclusions:

  • PhotoBioModulation with blue LED light demonstrates potential as a minimally-invasive approach for managing keloids and hypertrophic scars.
  • PBM may offer a novel therapeutic strategy when used in conjunction with existing treatments.
