Nucleic Acid Structure01:25

Nucleic Acid Structure

6.3K
The pentose sugar in DNA is deoxyribose, while in RNA the pentose sugar is ribose. The difference between the sugars is the presence of the hydroxyl group on the ribose's second carbon and a hydrogen on the deoxyribose's second carbon. The phosphate residue attaches to the hydroxyl group of the 5′ carbon of one sugar and the hydroxyl group of the 3′ carbon of the sugar of the next nucleotide, which forms  a 5′ to 3′ phosphodiester linkage.

Area of Science:

  • Structural Biology
  • Biophysics
  • Bioinformatics

Background:

  • Circular Dichroism (CD) spectroscopy is crucial for analyzing nucleic acid structure.
  • Existing data repositories lack comprehensive, curated CD spectral data for nucleic acids.
  • Synchrotron Radiation Circular Dichroism (SRCD) provides enhanced spectral resolution.

Purpose of the Study:

  • To establish a centralized, public repository for nucleic acid CD and SRCD spectral data.
  • To provide access to associated metadata, 3D structural models, and literature links.
  • To ensure data quality through expert validation and curation.

Main Methods:

  • Archiving and free distribution of CD and SRCD spectral data for various nucleic acids (DNA, RNA, hybrids, derivatives).
  • Integration of experimental metadata, primary sequence, and 3D structure models.
  • Implementation of expert validation and curation protocols for data quality assurance.

Main Results:

  • The Nucleic Acid Circular Dichroism Database (NACDDB) is now publicly accessible.
  • NACDDB contains a diverse collection of validated CD/SRCD spectral data linked to structural and literature information.
  • The database supports data submission, fostering community contribution.

Conclusions:

  • NACDDB serves as a valuable resource for researchers studying nucleic acid structure and dynamics.
  • The curated and validated data promotes reproducible research in the field.
  • The repository facilitates the integration of spectral data with structural and sequence information.
