Immunofluorescence Microscopy01:12

Immunofluorescence Microscopy

10.5K
A fluorescence microscope uses fluorescent chromophores called fluorochromes, which can absorb energy from a light source and then emit this energy as visible light. Fluorochromes include naturally fluorescent substances (such as chlorophylls) and fluorescent stains that are added to the specimen to create contrast. Dyes such as Texas red and FITC are examples of fluorochromes. Other examples include the nucleic acid dyes 4’,6’-diamidino-2-phenylindole (DAPI), and acridine orange.

The JAK-STAT Signaling Pathway01:20

The JAK-STAT Signaling Pathway

8.9K
Several cytokine receptors have tightly bound Janus kinase or JAK proteins attached at their cytosolic tail. Small signaling molecules such as cytokines, growth hormones, or prolactins bind to the cytokine receptors and initiate their dimerization. The dimerization brings the cytosolic JAKs together that trans-phosphorylate and activates each other. The activated JAKs now phosphorylate cytosolic tails of the cytokine receptors, which serve as binding sites for adaptor proteins such as  SH2...

Summary

This study reveals that drug concentrations in blood plasma do not accurately reflect drug amounts at target sites. Azalide macrolides concentrate in lung and gut epithelia, suggesting higher efficacy in mucous membranes than plasma levels indicate.

Area of Science:

  • Pharmacology
  • Biophysics
  • Cell Biology

Background:

  • Understanding drug distribution at target sites is crucial for accurate efficacy assessment.
  • Apparent drug concentrations in blood and plasma may not correlate with actual drug levels at the site of action.
  • Investigating the relationship between compound properties and organ/subcellular distribution is essential.

Purpose of the Study:

  • To investigate the relationship between physical properties, apparent bulk concentration, and organ/subcellular distribution of small molecules.
  • To determine if plasma or whole-blood drug concentrations underestimate levels in specific tissues.
  • To explore the distribution patterns of fluorescent JAK3 ligands and their macrolide conjugates.

Main Methods:

  • Utilized inherently fluorescent JAK3 ligands and macrolide conjugates.
  • Performed in vitro uptake studies into immune cells.
  • Analyzed murine pharmacokinetic samples using mass spectrometry and cryofluorescence microscopy.
  • Examined organ and subcellular distribution without fixation artifacts.

Main Results:

  • In vitro studies indicated substance association with granules or organelles in immune cells.
  • Cryofluorescence microscopy confirmed in vivo uptake of granules, consistent with in vitro observations.
  • Distribution patterns of coumarin-fluorophore and JAK3 inhibitor-carrying macrolides were similar.
  • Azalide macrolides showed a propensity to concentrate in lung and gut epithelia.
  • Plasma/whole-blood estimates likely underestimate macrolide concentrations in mucous membranes.

Conclusions:

  • Drug distribution is significantly influenced by the properties of larger molecules like macrolides.
  • Azalide macrolides accumulate in lung and gut epithelia, particularly mucous membranes.
  • Observed higher drug levels in barrier tissues may explain efficacy at sub-bacteriostatic doses.
  • Current plasma/blood concentration estimates may not accurately represent drug levels in these critical areas.
