The JAK-STAT Signaling Pathway01:20

The JAK-STAT Signaling Pathway

8.9K
Several cytokine receptors have tightly bound Janus kinase or JAK proteins attached at their cytosolic tail. Small signaling molecules such as cytokines, growth hormones, or prolactins bind to the cytokine receptors and initiate their dimerization. The dimerization brings the cytosolic JAKs together that trans-phosphorylate and activates each other. The activated JAKs now phosphorylate cytosolic tails of the cytokine receptors, which serve as binding sites for adaptor proteins such as  SH2...

Area of Science:

  • Immunology
  • Rheumatology
  • Preclinical Research

Background:

  • Inflammatory arthritides, like rheumatoid arthritis, are significant causes of disability.
  • Pre-clinical murine models are vital for identifying and validating therapeutic targets and drug candidates.
  • Lack of standardized scoring in animal models complicates result interpretation and comparison.

Purpose of the Study:

  • To introduce a detailed and implementable arthritis scoring system for murine models.
  • To enhance the capture of arthritis characteristics and disease progression monitoring.
  • To address ethical considerations and the principles of animal use reduction in arthritis research.

Main Methods:

  • Development and implementation of a comprehensive arthritis scoring system for experimental arthritis.
  • Detailed description of the scoring system for increased breadth of captured characteristics.
  • Inclusion of ethical and experimental design factors for animal research.

Main Results:

  • The proposed scoring system increases the breadth of captured arthritis characteristics.
  • It supports responsive and adaptive monitoring of disease progression in murine models.
  • Provides a framework for improved consistency and comparability across studies.

Conclusions:

  • Standardized scoring is essential for robust interpretation and comparison of results from murine arthritis models.
  • The developed system facilitates more reliable preclinical evaluation of novel therapeutics.
  • Emphasizes the importance of ethical considerations and the 3Rs (Replacement, Reduction, Refinement) in animal research.
