Background:

  • Oxytocin (OT), a neuropeptide, is known for its roles in social behavior and has demonstrated antinociceptive effects.
  • Previous human studies show intravenous OT reduces visceral pain and subcutaneous OT reduces postsurgical pain, but local effects on experimental pain were unstudied.

Purpose of the Study:

  • To investigate the local effects of subcutaneous oxytocin (OT) on experimental pain perception in healthy adults.
  • To determine if localized OT administration can modulate acute mechanical, pressure, and heat pain.

Main Methods:

  • A randomized, double-blind, placebo-controlled crossover study involving 18 healthy adults.
  • Participants received subcutaneous injections of synthetic OT (4 mcg) or saline placebo in a crossover design.
  • Pain perception (intensity, unpleasantness, threshold) for mechanical, pressure, and heat stimuli was assessed 10 minutes post-injection.

Main Results:

  • Subcutaneous OT significantly reduced subjective ratings of heat pain intensity and unpleasantness (P < .01).

Conclusions:

  • Subcutaneous oxytocin (OT) can modulate nociceptive processes in cutaneous tissues, reducing heat pain perception locally in humans.
  • Findings support OT's potential as a nonaddictive, localized treatment for acute pain, warranting further mechanistic and clinical research.
