Enhancer of zeste homolog 2 (EZH2) promotes multiple myeloma progression by activating the signal transducers and activators of transcription 3 (STAT3) pathway. Targeting EZH2 and STAT3 may offer new therapeutic strategies for multiple myeloma treatment.
Area of Science:
- Oncology
- Molecular Biology
- Biochemistry
Background:
- Multiple myeloma (MM) is a bone marrow malignancy characterized by clonal plasma cell proliferation.
- The enhancer of zeste homolog 2 (EZH2) and signal transducers and activators of transcription 3 (STAT3) pathway are implicated in various malignancies.
Purpose of the Study:
- To investigate the role of EZH2 in multiple myeloma (MM) progression.
- To elucidate the involvement of the STAT3 signaling pathway in EZH2-mediated MM pathogenesis.
- To identify potential therapeutic targets for MM.
Main Methods:
- Quantitative real-time PCR and western blot to assess EZH2 expression in MM cell lines.
- In vitro manipulation of EZH2 and STAT3 activation using transfection, colivelin, and stattic.
- Functional assays including colony formation, transwell migration, and flow cytometry to evaluate cell proliferation, migration, and apoptosis.
Main Results:
- EZH2 was significantly overexpressed in multiple MM cell lines at both mRNA and protein levels.
- Increased EZH2 expression enhanced MM cell proliferation and migration while inhibiting apoptosis.
- These effects were reversed by STAT3 inhibition and mimicked by STAT3 activation, indicating EZH2 acts via the STAT3 pathway.
Conclusions:
- EZH2 promotes multiple myeloma progression by activating the STAT3 pathway.
- Both EZH2 and STAT3 pathways represent promising therapeutic targets for multiple myeloma treatment.
