This study introduces a novel electrochemical aptamer-based biosensor using oxazine ATTO 700. This innovation enables true multiplex, continuous, and real-time measurement of two molecules in undiluted whole blood on a single electrode.
Area of Science:
- Electrochemistry
- Biosensor Technology
- Molecular Diagnostics
Background:
- Multiplex molecular measurements using electrochemical biosensors often require multiple electrodes, posing challenges for specific patient populations like neonates and hemophiliacs.
- Existing methods are not ideal for continuous, real-time analysis in complex biological samples such as whole blood.
Purpose of the Study:
- To develop a single-electrode electrochemical aptamer-based biosensor capable of true multiplex detection.
- To enable continuous and real-time measurement of two distinct molecules simultaneously in undiluted whole blood.
Main Methods:
- Incorporation of the oxazine ATTO 700 dye into electrochemical aptamer-based sensor design.
- Testing the sensor's performance for simultaneous detection of two target molecules in undiluted whole blood samples.
Main Results:
- Demonstrated the feasibility of achieving "true" multiplex detection using a single electrode.
- Successfully performed continuous and real-time measurements of two different molecules in undiluted whole blood.
Conclusions:
- The developed oxazine ATTO 700-based electrochemical aptamer biosensor offers a simplified approach for multiplex analysis.
- This single-electrode system provides a promising platform for real-time diagnostics in challenging biological matrices, overcoming limitations of multi-electrode systems.
