Area of Science:
- Cell biology
- Bioimaging
- Software development
Background:
- Video-microscopy is crucial for single-cell behavior analysis.
- Manual tracking of real-time cellular events is time-consuming and prone to error.
- AI-driven analysis is emerging but manual intervention is still required for some events.
Purpose of the Study:
- To develop a user-friendly software, Toto-Cell, for analyzing video-microscopy data.
- To enable quantification of previously difficult-to-measure cellular parameters.
- To facilitate the analysis of cellular events induced by various treatments.
Main Methods:
- Developed Toto-Cell software for video-microscopy data analysis.
- Utilized HEC-1-A endometrial cell line for experiments.
- Treated cells with cisplatin, doxorubicin, and AZD5363 (Akt inhibitor).
- Tracked parameters: mitosis (normal/abnormal), cell death/fusion ratios, and cell cycle length.
Main Results:
- Toto-Cell successfully quantified various cellular parameters.
- Demonstrated significant heterogeneity in cellular responses within single treatments.
- Highlighted previously challenging parameters like mitosis and cell death ratios.
Conclusions:
- Toto-Cell is an effective tool for quantitative and qualitative analysis of video-microscopy data.
- The software enhances accuracy compared to manual calculations.
- It aids researchers in understanding complex cellular dynamics and treatment effects.

Protein Dynamics in Living Cells
Different fluorescence-based techniques are used to study the protein dynamics in living cells. These techniques include FRAP, FRET, and PET.
Fluorescent recovery after photobleaching (FRAP) is a fluorescent-protein-based detection technique used to quantify protein movement rates within the cell. This method exposes a small portion of the cell to an intense laser beam. The laser beam causes permanent photobleaching of the fluorophore-tagged proteins in the exposed region. As the bleached...

Total Internal Reflection Fluorescence Microscopy
Total internal reflection fluorescence microscopy or TIRF is an advanced microscopic technique used to visualize fluorophores in samples close to a solid surface with a higher refractive index, such as a glass coverslip. TIRF only allows fluorophores in proximity to the solid surface to be excited. When light from a medium with a lower refractive index (such as air) hits the glass coverslip at a critical angle, the light undergoes total internal reflection stead of passing through the glass.
