A new method rapidly detects Epidermal Growth Factor Receptor (EGFR) mutations in lung cancer. This Thermus thermophilus Argonaute (Ttago)-Coupled-Multiplex-digital-recombinase polymerase amplification (RPA)-CRISPR/Cas12a (TCMDC) assay offers a fast, sensitive tool for non-small cell lung cancer (NSCLC) diagnosis.
Area of Science:
- Biotechnology
- Molecular Diagnostics
- Oncology
Background:
- Epidermal Growth Factor Receptor (EGFR) is a critical target in non-small cell lung cancer (NSCLC) management.
- Existing EGFR detection methods face challenges with long detection times and false positives.
- Developing rapid, sensitive, and specific diagnostic tools is crucial for improving NSCLC patient outcomes.
Purpose of the Study:
- To develop a novel, rapid, and highly sensitive detection method for EGFR mutations in NSCLC.
- To address the limitations of current EGFR detection technologies in terms of speed and accuracy.
- To provide a potential clinical tool for early diagnosis and treatment monitoring of lung cancer.
Main Methods:
- Development of a Thermus thermophilus Argonaute (Ttago)-Coupled-Multiplex-digital-recombinase polymerase amplification (RPA)-Clustered regularly interspaced short palindromic repeats (CRISPR)/Cas12a (TCMDC) assay.
- Integration of EGFR mutation template enrichment using TtAgo and double-guide DNA.
- Utilization of PAM-site-independent CRISPR RNAs targeting EGFR genes combined with RPA and CRISPR-Cas12a in a single pot.
Main Results:
- The TCMDC method achieved ultra-sensitive detection of EGFR mutations (down to 1 copy, 0.1%).
- The assay demonstrated visual detectability and high specificity.
- Successful validation using clinical serum samples from lung cancer patients, with detection completed within 1 hour.
Conclusions:
- The developed TCMDC method offers a rapid, sensitive, and visually detectable approach for EGFR mutation detection.
- This assay presents a promising and efficient clinical tool for lung cancer diagnosis and management.
- The method overcomes limitations of traditional techniques, potentially enhancing early detection and treatment strategies for NSCLC.
