Adding a hypoxia-mimicking compound (IOX2) to transforming growth factor beta 1 (TGFβ1)-stimulated hepatic stellate cells (HSCs) improved the preclinical model of liver fibrosis. This approach enhances the accuracy of in vitro models for studying fibrosis.
Area of Science:
- Biomedical research
- Cell biology
- Translational medicine
Background:
- Preclinical models of liver fibrosis incompletely replicate human disease.
- Traditional hepatic stellate cell (HSC) models lack crucial hypoxia-induced pathways.
Purpose of the Study:
- To investigate if a hypoxia-mimicking compound (IOX2) can improve the accuracy of HSC models for liver fibrosis.
- To assess the functional fibrotic phenotype in vitro.
Main Methods:
- Human primary HSCs were stimulated with transforming growth factor beta 1 (TGFβ1) with or without IOX2.
- Fibrotic markers (collagen, TIMP-1, Fibronectin) and cell viability were measured.
- Next-generation sequencing (NGS) and pathway analysis (IPA) were used for mechanistic insights.
Main Results:
- TGFβ1 + IOX2 significantly elevated fibrotic marker levels.
- Hypoxia-related genes and collagen modifications increased dose-dependently.
- The IOX2-treated model showed improved representation of human fibrotic gene expression.
Conclusions:
- IOX2 administration enhances the ability of HSC models to recapitulate liver fibrosis.
- Hypoxia-mimicking compounds can improve the translational value of in vitro fibrosis models.
- This approach offers valuable insights into liver fibrosis pathogenesis and therapeutic strategies.
