Abstract:
Graphene family materials possess significant application potential in the fabrication of peripheral nerve repair materials; however, optimization remains necessary for developing neural repair materials with superior biocompatibility. Graphene oxide quantum dots (GOQDs) exhibit enhanced biocompatibility while maintaining the neural repair efficacy characteristic of graphene family materials. Previous studies have reported the interactions and mechanisms of GOQDs with macrophages and vascular endothelial cells during peripheral nerve repair processes, yet their interactions and mechanisms with Schwann cells in peripheral nerve regeneration remain unclear. To address this knowledge gap, this study employed electrospinning technology to fabricate GOQDs-loaded PLGA (poly(lactic-co-glycolic acid)) nerve scaffolds. Our findings demonstrate that GOQDs incorporation not only enhances the mechanical properties and hydrophilicity of the composite nanofiber scaffolds but also promotes Schwann cell proliferation. Furthermore, RNA sequencing analysis revealed that GOQD-mediated Schwann cell proliferation is regulated through the Wnt signaling pathway. This study provides theoretical foundations and experimental evidence supporting the clinical translation of graphene family materials as nerve repair biomaterials.
