An asymmetric total synthesis of chondrochloren A, a polyketide-derived antibiotic characterized by an unusual Z-enamide attached to a chloro-hydroxy-styryl moiety and a densely oxygenated central segment, is described. A Paterson aldol reaction on an aldehyde generated from d-glucose provided a key amide fragment. Subsequent coupling of this intermediate with a Z-vinyl iodide, followed by protecting group removal, furnished the fully assembled natural product.
