Construction of Fused Bisindoles through Catalytic Enantioselective Intramolecular Povarov Reaction

Abstract:
An enantioselective intramolecular Povarov reaction was established to construct 1,7-annulated hexacyclic bisindoles. With newly designed 7-formalindole-tethered alkenes, this one-pot, metal-free process occurred quickly to deliver the desired structures as a single diastereomer in 35-98% yield and 61-98% ee. This method provides direct access to a distinctive double-ring-fused bisindole architecture, offering a novel approach to complex polycyclic bisindole systems.
