MinION Sequencer Demonstrates Strong DNA Sequencing Accuracy and Identity

Abstract:
Nanopore sequencing with the MinION device has revolutionized genomic research. This study evaluated the MinION nanopore sequencer for accuracy, reliability, and reusability. Sequencing of λ phage DNA and E. coli DNA achieved high accuracy and identity levels of at least 90%. Results were comparable to previous studies, confirming the MinION's ability to capture genomic sequences larger than 4.6 Mbp. Decreased accuracy and identity with repeated nanopore use were observed, indicating the need for caution when comparing runs from the same flow cell.
