Dynamic Fourier-domain full-field optical coherence microscopy
Abstract:
Fourier-domain Full-Field Optical Coherence Microscopy (FD-FF-OCM) enables high-resolution, high-speed volumetric imaging but provides limited intrinsic contrast. We present dynamic FD-FF-OCM, which enhances contrast by analyzing temporal fluctuations in the complex FD-FF-OCM signal. The system provides access to fluctuation frequencies of 0.3-58 Hz while acquiring wide-field volumetric data over a 200 µm axial range in 3.3 s, with dynamic cellular features clearly resolved down to 70 µm and blurred structural contrast remaining visible at larger depths. Imaging of freshly excised mouse liver demonstrates substantially improved contrast over static FD-FF-OCM, enabling clearer visualization of hepatocytes, their nuclei, and sinusoids.

Phase Contrast and Differential Interference Contrast Microscopy
In-phase-contrast microscopes, interference between light directly passing through a cell and light refracted by cellular components is used to create high-contrast, high-resolution images without staining. It is the oldest and simplest type of microscope that creates an image by altering the wavelengths of light rays passing through the specimen. Altered wavelength paths are created using an annular stop in the condenser. The annular stop produces a hollow cone of...
