Abstract:
An innovative high-performance thin-layer chromatography approach with second-order spectral detection (HPTLC-SOS) integrates multi-wavelength UV-Vis data (286-525 nm) across eight discrete retardation factor (Rf) positions to characterize complex oenological matrices. Seventy-one Argentine wines from three regions and multiple monovarietals (including Malbec and Cabernet Sauvignon) were analyzed. Principal Component Analysis (PCA) of this compact three-dimensional dataset achieved prominent separation based simultaneously on varietal identity (PC1) and geographical origin (PC2). Cluster-specific loading heatmaps validated the chemical basis of this discrimination. Furthermore, HPTLC-SOS outperformed a reference HPLC-DAD technique in throughput (7.2 vs. 60 min/sample) and greenness, as confirmed by the AGREE metric (0.74 vs. 0.50). This superiority stems from parallel processing, passive capillary development, and over 98% reduction in solvent waste. Consequently, this discrete-sampling strategy represents a reliable, high-throughput, and sustainable analytical platform for wine traceability and quality control.
