Abstract:
A practical total synthesis of streptospherin A (1), a cancer stem cell inhibitor, is reported. The key intermediates are obtained stereoselectively in a concise synthesis by a combination of asymmetric allylation and hydroxy-directed stereoselective diboration. These intermediates were coupled via N-acetylcysteamine thioester-mediated Liebeskind-Srogl cross-coupling to achieve the total synthesis of 1.
