Abstract:
Objective: To establish a method for the determination of resorcinol in serum by liquid chromatography tandem mass spectrometry. Methods: In June 2024, the serum samples were extracted with ammoniated acetonitrile, cleaned up by a PRiME MCX solid phase extraction column. Separation was performed on a Waters HSS T3 column (100 mm×2.1 mm, 1.8 μm) with a linear gradient elution program of water and acetonitrile as the mobile phase. The analyte was measured by using electrospray ion (ESI) in the negative multiple reaction monitoring (MRM) mode. Quantification was carried out by the external standard method. Results: A good linearity was obtained in the concentration range of 2.0~100.0 μg/L for target compound with the correlation coefficient greater than 0.999. The limit of detection (LOD) and the limit of quantitation (LOQ) were 0.7 μg/L and 2.0 μg/L.The average spiked recovery was 89.8%, the intra-day precision was 2.8%~5.6%, and the inter-day precision was 4.1%~7.3%. Conclusion: The established method was of high accuracy and sensitivity, and was suitable for the determination of resorcinol in human serum.
