    Background:

    • Epithelioid sarcoma is a rare soft tissue tumor with uncertain origins.
    • Previous studies identified specific ultrastructural features of epithelioid sarcoma cells.

    Observation:

    • Electron microscopy revealed abundant 10 nm cytoplasmic filaments, desmosome-like junctions, and cystic spaces in tumor cells.
    • Unique findings included 10 nm filaments with condensed, whorled patterns and 5'-nucleotidase activity in tumor cell membranes.

    Findings:

    • Tumor cells exhibited 5'-nucleotidase activity, a marker also found in synovial and endothelial cells.
    • Unlike endothelial cells, both tumor and synovial cells lacked adenosine triphosphatase activity.

    Implications:

    • The observed enzyme activity patterns support the hypothesis that epithelioid sarcoma originates from mesenchymal cells differentiating towards synovial cells.
    • This research contributes to understanding the histogenesis of epithelioid sarcoma and its relationship with synovial tissues.
