    Background:

    • Fetal Alcohol Syndrome (FAS) is a pattern of malformations observed in offspring of women with chronic alcohol consumption.
    • Animal models are crucial for investigating the mechanisms underlying FAS.
    • Previous research has focused on establishing and studying these animal models.

    Purpose of the Study:

    • To investigate the teratogenic effects of a single alcohol exposure during gestation in Sprague-Dawley rats.
    • To identify specific malformations induced by prenatal alcohol administration.
    • To examine the incidence of cardiovascular defects in the offspring.

    Main Methods:

    • Pregnant Sprague-Dawley rats received a single intraperitoneal injection of ethanol on day 10 of gestation.
    • Fetuses were collected on day 18 of gestation for examination.
    • External malformations were documented, and internal tissues were histologically processed (Bouin's fixative, hematoxylin and eosin staining) for detailed analysis.

    Main Results:

    • External malformations like adactyly, wrinkled skin, and gastroschisis were infrequent.
    • Histological examination revealed severe umbilical hernias in experimental fetuses.
    • A significant incidence (41%) of ostium secundum type interatrial-septal defects was observed in the offspring of alcohol-exposed dams, absent in controls.

    Conclusions:

    • A single prenatal alcohol exposure can induce significant developmental abnormalities in rat fetuses.
    • The study identified a high prevalence of specific heart defects, particularly ostium secundum atrial septal defects, following gestational alcohol exposure.
    • These findings underscore the teratogenic potential of alcohol and its impact on fetal cardiovascular development.
